摘要
利用RT-PCR技术从宁夏枸杞(Lycium barbarumL.)果实cDNA中分离出八氢番茄红素合成酶(Phytoene syn-thase,PSY)和番茄红素β环化酶(Lycopeneβcyclase,LCYB)同源基因cDNA片段,分别命名为Lyb-Psy和Lyb-Lcyb.测序结果表明,Lyb-Psy序列长402 bp,编码134个氨基酸;Lyb-Lcyb序列长555 bp,编码184个氨基酸.BLAST结果显示,Lyb-Psy和Lyb-Lcyb所推导的氨基酸与烟草、番茄对应序列同源性最高,分别为96%和92%,与其他物种对应区域也有86%~90%的序列同源性.
The cDNA sequences for phytoene synthase(PSY) and lycopene β cyclase(LCYB) were cloned from fruits of wolfberry by RT-PCR,which were designated as Lyb-Psy and Lyb-Lcyb respectively.Sequencing results revealed that Lyb-Psy was 402 bp in length,encoding 134 amino acids,while Lyb-Lcyb was 555 bp in length,encoding 184 amino acids.BLAST results showed that the deduced amino acids of Lyb-Psy and Lyb-Lcyb shared 96% and 92% identities with those of Nicotiana tabacum and Solanum lycopersicum,as well as 86%~90% identities with corresponding regions of other plants.Our present results could provide theoretical basis on exploring the carotenoids metabolism in wolfberry.
