摘要
目的探讨染料木素对光动力疗法杀伤K562细胞的影响。方法通过检测细胞数目变化情况、细胞膜的氧化损伤以及细胞凋亡进行研究。结果染料木素可以抑制K562细胞的生长,增加光动力疗法对K562细胞的杀伤(24 h孵育后)。结论染料木素同光动力疗法有协同作用。抗氧化剂对光动力疗法的影响很可能决定于其对单线态氧产生的影响。提示染料木素同光动力疗法联合使用会有利于癌症的治疗。
Aim To investigate the influence of genistein on Photodynamic therapy (PDT). Methods The hemacytometer was applied to observe cell proliferation. Cell apoptosis was determined by flow cytometry. Lipid peroxidation was evaluated by determining the malondialdehyde (MDA) amount using fluorophotometry. Results Genistein inhibits K562 cells proliferation. It enhanced cells death after incubating 24h in a dose-dependent manner when undergoing PDT treatment. Genistein supplementation increased the amount of MDA and cell apoptosis in ALA-PDT. Conclusion Genistein exhibited a synergistic effect on PDT. The effect of antioxidants on PDT may be determined by its sensitization ability to singlet oxygen. The research implied that genistein and PDT combined may improve the effectiveness of cancer therapy.
