摘要
目的研究骨髓来源的间充质干细胞对胃癌细胞SGC-7901增殖的影响,为胃癌的治疗与研究提供新的思路及方法。方法原代培养骨髓来源的间充质干细胞,传代培养胃癌细胞SGC-7901,间充质干细胞及5-Fu对胃癌细胞SGC-7901增殖能力的影响采用MTT比色法。结果 24 h内,与MCM组及DMEM相比,MSC组胃癌细胞SGC-7901的吸光度降低较显著(P<0.05);与DMEM相比,24 h后MCM组胃癌细胞SGC-7901吸光度开始降低,该现象可延长至72 h(P<0.05)。加入5-Fu后,与DMEM组相比2,4 h后MCM组SGC-7901吸光度降低较显著(P<0.05);但随着时间的延长至72 h,两组间的差异消失(P>0.05)。结论骨髓来源的间充质干细胞可抑制胃癌细胞SGC-7901的增殖,并协同5-Fu对胃癌细胞SGC7-901的增殖起抑制作用。
Objective To study the effect of bone marrow-derived mesenchymal stem cells on proliferation of gastric carcinoma cell SGC-7901, and to provide new ideas and methods for treatment and research of gastric carcinoma. Methods Bone marrow-derived mesenchymal stem cells were primary cultured, gastric carcinoma cell SGC-7901 were subcultured, and an interaction model of mesenchymal stem cells and gastric carcinoma cell SGC-7901 in vitro was established. The effect of bone marrow-derived mesenchymal stem cells on proliferation of gastric carcinoma cell SGC-7901 was analyzed by MTT. Results Compared with MCM group an.d DMEM group, the OD value of gastric carcinoma cell SGC-7901 was lower in MSC group(P 〈 0.05 ) ; compared with DMEM group, the OD value of gastric carcinoma cell SGC-7901 was reduced after 24 h in MCM group (P 〈 0.05). After 5-Fu was added, compared with DMEM group, the OD value of gastric carcinoma cell SGC-7901 was reduced after 24 h in MCM group ( P 〈 0.05 ), and the difference was disappeared in 72 h ( P 〉 0.05 ). Conclusion Bone marrow-derived mesenchymal stem cells can inhibit proliferation of gastric carcinoma cell SGC- 7901 in vitro,and the inhibition is synergistic with 5-Fu.
