摘要
[目的]探讨有机磷化合物(organophosphorvs compounds,OPs)引起迟发型神经毒性(organophosphorus ester-induced delayed neuropathy,OPIDN)后鸡的神经组织中髓鞘碱性蛋白(myelin basic prote in,MBP)表达的变化。[方法]罗曼鹤母鸡48只,随机分成染毒组:给予1000 mg/kg三磷甲苯磷酸酯(tri-ortho-cresyl phosphate,TOCP)、苯甲基磺酰氟(phenylm ethanesu l fonyl fluoride,PMSF)前干预组:在染毒组之前给予40 mg/kg PMSF和对照组(生理盐水),每组16只。染毒第5天和第20天,每组分别处死8只动物,低温环境下分离腰髓,提取总蛋白。用双向电泳和质谱分析技术,筛选可能与脱髓鞘相关的差异表达蛋白。[结果]经双向电泳和质谱分析表明,在染毒第5天TOCP组及PMSF前干预组MBP变化与对照组比较,差异无显著性意义;第20天与对照组比较,TOCP组与PMSF前干预组的MBP表达分别为0.3667与0.7601(比值在0.5~2之间无意义),前者表达下调达2.7倍,差异有显著性意义,后者差异无显著性意义。[结论]TOCP暴露诱导鸡脊髓神经组织MBP表达显著下调,其变化可能与OPIDN模型动物神经组织的脱髓鞘现象有关。
[Objective] To investigate the special difference of myelin basic protein in the hen spinal cord tissue after organophosphorus ester-induced delayed neuropathy.[Methods] Forty-eight Roman hens were randomly divided into three groups: treated-group(1000 mg/kg tri-ortho-cresyl phosphate,TOCP);intervented-group(40 mg/kg phenylmethanesulfonyl fluoride(PMSF) before treated-group;control group(tap water),every group have 16 hens.Eight hens in every groups were sacrificed on the d5、d20 separately,which separate spinal cord tissue at low temperatures and extract total protein.With solid phase PH gradient SDS-PAGE two-dimensional electrophoresis and mass spectrometry,we screen differently expressed proteins related to demyelination.[Results] Comparing with control group,the difference of myelin basic protein(MBP) has no significance on the d5,while on the d20,the expression rates treated-group、intervented-group were 0.3667 and 0.7601 receptively(ratio have no significace beween 0.5 and 2).[Conclusion] The expression of MBP is downregulated in the hen spinal cord tissue after organophosphorus ester-induced delay neuropathy,which may relate to demyelination of OPIDN model.
