G3BP和OPN在喉鳞状细胞癌的表达及其临床意义

Clinical implications of G3BP and OPN expression in laryngeal squamous cell carcinomaa

目的研究喉鳞癌组织中GTP酶激活蛋白SH3功能区结合蛋白(G3BP)和骨桥蛋白(OPN)的表达及其与喉鳞癌生物学行为的关系。方法 91例喉鳞癌组织标本,应用免疫组织化学方法检测G3BP和OPN表达水平,分析检测结果与瘤细胞分化程度、TNM分期、淋巴结转移和预后的关系。结果 G3BP阳性率为84.7%,其表达水平与瘤细胞分化程度(P=0.008)、临床分期(P=0.005)、淋巴结转移(P=0.000)及术后肿瘤复发与转移(P=0.000)均密切相关;G3BP表达水平越高,患者术后生存时间越短(P=0.000)。OPN阳性率为76.5%,其表达水平与临床分期(P=0.035)、淋巴结转移(P=0.005)及术后肿瘤复发与转移(P=0.017)密切相关,而与瘤细胞分化程度(P=0.211)无关;OPN表达水平越高,患者术后生存时间越短(P=0.032)。G3BP与OPN的表达水平之间存在正相关(rs=0.556,P=0.000)。结论 G3BP与OPN表达水平可作为判断喉鳞癌患者预后的有效参考指标。

Objective To investigate the implications of Ras-GTPase-activating protein SH 3 domain binding protein （G3BP） and osteopontin protein （OPN） in laryngeal squamous cell carcinoma （LSCC） based on a clinical trial. Methods Included in this study were 91 cases with LSCC, with their tissue samples taken from surgically removed focus to determine the expressive levels of G3BP and OPN proteins by immunohistochemistry. Then, a comparative study was made to analyze the association of expressive levels in these two kinds of protein with differentiation degree of tumor cells, TNM staging, lymph node metastasis and prognosis of patients with this lesion. Resuts The positive expressive rate of G3BP was 84.7% in this group of cases, with very higher correlation of G3BP expressive level with the differentiating degree of tumor cells （P=0.008）, clinical staging （P=0.005）, lymphatic metastasis （P=0.000） and post-operative tumor recurrence and/or distant metastasis （P=0.000）. In the other hand, the positive expression rate of OPN was 76.5% among these samples, with significant correlation in its expressive level with clinical staging （P=0.035）, lymphatic metastasis （P=0.005） and post-operative tumor recurrence and/or distant metastasis （P=0.017） as well, while no significant relation with the differentiating degree of tumor cells （P=0.211） found. It was shown from this study that the expressive levels of G3BP and OPN were both significantly related to the survival period of LSCC patients in a negative correlation way （P=-0.000 and P=0.032 respectively for them） Furthermore, positively correlated with each other were the expressive levels of G3BP and OPN in this group of LSCC patients （r=0.556, P=0.000）. Conclusions The expressive levels of G3BP and OPN can be effectively taken as markers of prognostic evaluation for overall survival of LSCC patients.
