RP-HPLC法测定板栗种仁中原儿茶酸、对羟基苯甲酸和异香草酸的含量 被引量:6

摘要 目的建立同时测定板栗种仁中原儿茶酸、对羟基苯甲酸、异香草酸含量的方法。方法采用Dikma Diamonsil C18柱(250mm×4.6mm,5um),以甲醇-体积分数为0.05%的甲酸水溶液为流动相进行梯度洗脱,流速为1.0mL·min^-1,检测波长为260nm,柱温为室温。结果原儿茶酸、对羟基苯甲酸、异香草酸质量浓度分别在10—100mg·L^-1、5~50mg·L^-1、50~500mg·L^-1内与峰面积具有良好的线性关系(r≥0.9998,n=6),方法平均回收率分别为98.5%、98.0%、99.6%。结论本方法可用于同时测定板栗种仁中原儿茶酸、对羟基苯甲酸、异香草酸的含量。 Objective To develop an RP-HPLC method for the simultaneous determination of the contents of protocatechuic acid,p-hydroxybenzoic acid and isovallic acid from seeds of Castanea moUissima Blume. Methods The analysis was carried out on a Dikma Diamonsil C18 (250 mm × 4. 6 mm, 5 μm)column;the mobile phase was composed of methanol and 0. 05% formic acid with gradient elution at a flow rate of 1.0 mL. min^-1. The detection wavelength was set at 254 nm and column temperature was room temperature. Results The linear ranges of protocatechuic acid,p-hydroxybenzoic acid and isovallic acid were 10- 100 mg. L^- 1,5- 50 mg. L^- 1,50- 500 mg. L^- 1 ( r≥ 0. 999 8, n = 6 ), respectively. The average recoveries were 98.5% ,98.0% ,99.6% ,respectively. Conclusions This method is simple and can be used to determine the contents of the above three components with satisfactory accuracy and reproducibility.
