摘要
应用RAPD技术对43个贺兰山紫蘑菇(Cortinarius)属菌株进行遗传多样性分析。结果表明,在供试的17条RAPD随机引物中,有7条可扩增出清晰、稳定的DNA条带,利用筛选出的7条引物进行RAPD扩增,共得到79条清晰的DNA条带,其中68条多态性片段占总扩增片段的86.1%;聚类分析表明,在相似系数0.63水平时,供试的43个菌株被分为紫红丝膜菌(C.rufo-olivaceus)和蓝丝膜菌((C.caerulescens)两个组群,同时紫红丝膜菌种内41个样品间的差异较显著,其中种内最大相似系数为0.956,最小相似系数为0.515;在相似性系数为0.68时,可将紫红丝膜菌的41个样品分为7类。研究结果表明RAPD技术可以有效地区分紫蘑菇菌株并分析种内的遗传多样性。
Genetic diversity among 43 strains of Cortinarius was analyzed by RAPD using seven primers selected from 17 test primers previously shown to display specific and stable amplification patterns.A total of 79 RAPD bands were detected,of which 68(86.1%) were polymorphic.A dendrogram was constructed using the amplification data that separated the strains into two groups at a coefficient value of 0.63.The two groups consisted of 41 C.rufo-olivaceus strains and two of C.caerulescens.The 41 C.rufo-olivaceus strains exhibited greater genetic diversity with a maximum similarity coefficient of 0.956 and a minimum similarity coefficient of 0.515.RAPD analysis separated the 41 strains into seven groups at a coefficient value of 0.68,and the method was therefore deemed effective in reflecting genetic diversity among members of this species.
