摘要
目的:在慢性粒细胞白血病病人(CML),CD3ζ链表达明显下降,分析与CD3ζ存在互补关系的FcεRⅠγ基因在CML患者中表达水平,以了解T细胞免疫中TCR信号转导的变化情况。方法:利用SYBR GreenⅠ实时荧光定量RT-PCR法测定CML患者和健康人各20例的外周血单个核细胞(PBMCs)的FcεRⅠγ基因表达水平,以β2微球蛋白基因(β2MG)作为内参照,采用相对定量公式:2-ΔCt×100%,计算FcεRⅠγ链的相对mRNA表达量。结果:CML患者PBMCs中FcεRⅠγ基因表达水平明显高于健康对照组(P<0.001)。FcεRⅠγ表达水平变化与病人外周血CD3+T细胞比例无相关性。结论:CML病人中FcεRⅠγ表达上升,提示FcεRⅠγ可能在一定程度调节CD3ζ链缺陷所带来的T细胞免疫异常。
Aim:In order to understand the change of TCR signal transduction in T-cell immunization,the expression level of FcεRⅠγ chain gene,which is complementary with the CD3ζ,was analyzed,based on the down expression of CD3ζ chain gene in patients with chronic myelocytic leukemia(CML).Methods: Real-time PCR with SYBR GreenⅠ technology was used to determine the expression level of FcεRⅠγ chain in peripheral blood mononuclear cells(PBMCs)from 20 patients with CML and 20 healthy individuals.β2-microglobulin gene(β2M) was used as an endogenous reference.Relative changes in FcεRⅠγ gene expression level were determined by the 2-△Ct method between patients with CML and normal individuals.Results: The results showed that the expression levels of FcεRⅠγ gene in PBMCs from CML patients were higher than that from healthy individuals(P 0.001).However,the expression levels of FcεRⅠγ gene had no correlation with the percentage of CD3+ T cell in PBMCs.Conclusion:Overexpression of FcεRⅠγ gene might partially overcome the abnormal T-cell immune state due to the deficiency of CD3ζ gene in patients with CML.
