HPLC梯度洗脱法测定山栀苷甲酯、8-O-乙酰山栀苷甲酯在独一味中含量 被引量:1

摘要 目的建立独一味中山栀苷甲酯、8-O-乙酰山栀苷甲酯的HPLC检测方法。方法色谱柱Dikma platisil C18(250 mm×4.6 mm,5μm),柱温35℃,以乙腈-水进行梯度洗脱,流速1.0 mL/min,检测波长235 nm。结果山栀苷甲酯进样量在0.023 0~ 3.450 0μg范围内呈良好线性关系,r=0.999 3,8-O-乙酰山栀苷甲酯进样量在0.041 82~ 6.273μg范围内呈良好线性关系,r=0.999 6。山栀苷甲酯平均回收率为99.79%,RSD为1.63%,8-O-乙酰山栀苷甲酯平均回收率为99.62%,RSD为1.79%。结论 HPLC测定独一味中山栀苷甲酯、8-O-乙酰山栀苷甲酯结果准确可靠,快速,灵敏,重现性好。 Objective To establish a method to determine Shanzhiside methyl ester and 8-O-acetylshanzhiside methyl ester in LamiophIomis rotate (Benth.) Kudo by HPLC. Methods HPLC analysis was performed on Dikma platisil C18 ( 250 mm×4.6 mm, 5 μm) column with gradient elution of acetonitrile-water at a flow rate of 1.0mL/min. The column temperature was 30℃and the detection wavelength was 235nm. Results The linear range of Shanzhiside methyl ester was 0.023 0-3.450 0 μg, r-=0.999 3. The linear range of 8-O-acetylshanzhiside methyl ester was 0.041 82-6.273 μg, r=0.999 6.The average recovery of Shanzhiside methyl ester was 99.79% with RSD 1.63%, the average recovery of 8-O-acetylshanzhiside methyl ester was 99.62% with RSD 1.79%. Conclusion The method is sentitive, rapid and reproducible.
