摘要
目的:探讨桂枝合地黄汤对更年期潮热大鼠尾部皮肤温度(tail skin temperature,TST)、血清雌二醇(E2)、促性腺激素释放激素(GnRH)、β-内啡肽(β-EP)水平的影响。方法:将成年雌性大鼠随机分为4组。除对照组采用假手术外,其它各组均手术摘除双侧卵巢复制更年期动物模型。治疗组于去势后中药灌胃。各组大鼠每周连续测量TST,9周后酶免法测各组血清E2、GnRH、β-EP含量。结果:与对照组和治疗组比较,20℃时模型组大鼠的尾温在去势后1~4周间出现明显升高(P<0.05);血清GnRH升高(P<0.05),β-EP显著下降(P<0.01)。当提高环境温度至30℃时,模型组尾温明显上升(P<0.05)。结论:补肾和营卫法可能通过调节去势雌性大鼠GnRH的合成与释放从而调节中枢神经递质β-EP等的水平以改善更年期潮热症状。
Objective :To study the effects of Cinnamon Twig with Six Drugs including Rehmannia Decoction on tail skin temperature ( TST), estradiol ( E2 ), gonadotropin releasing hormone (GnRH) andβ- endorphin ( β - EP ) in menopa- usal hot flushes rats. Methods: Adult Wistar female rats were randomly divided into four groups:normal group, model group, Chinese medicine high - and low - dose groups. Except the rats in normal group, the rats received removal of bi- lateral ovaries to establish menopausal rat models. All of the TST were measured and its mean values were calculated every week from first week before till 9th week after the ovariectomy. Chinese medicine group were given traditional herbal decoction by ig administration for 9 weeks. The serum E2, GnRH, β - EP levels were measured each group by enzyme - linked immunosorbent assay in the end. Result The TST of model group was elevated( P 〈 0.05) from the first till 4th week after the ovariectomy compared with that of other groups at 20℃ and it Was increased( P 〈0.05) from 5th till 9th week after the elevation of ambient temperature at 30℃. The level of GnRH was increased( P 〈 0.05 ) and the β - EP was decreased( P 〈0.01 ) in the model group compared with those in other groups in the end. The level of serum E2was decreased( P 〈0.01 or P 〈0.05) in model group and in Chinese medicine group compared with that in normal group. Conclusion the role of TCM Treatment of Reinforcing Kidney and Regulating Ying - wei was not to regulate the level of serum E2but to modulate the secretion of serum GnRH and β - EP in menopausal hot flushes rats.
