温补肾阳方对抑郁症大鼠行为学及脑内单胺递质影响研究

Study on the Effect of Prescription for Warming and Recuperating the Kidney-yang on the Behavior and Monoamine Neurotransmitters in the Depression Rat

目的:探讨温补肾阳方对抑郁模型大鼠行为学及脑内单胺递质的影响。方法:选择体重和旷野试验(open-field test)评分相近的72只SD大鼠,随机分为空白组,模型组,温补肾阳方低、中、高剂量组,逍遥散大剂量组,应用分养和长期不可预知的中等强度应激造成大鼠抑郁模型,定期测定各组大鼠行为学,采用酶联接免疫吸附剂(ELISA)法检测大鼠5-羟色胺(5-HT)、多巴胺(DA)、去甲肾上腺素(NE)含量,观察温补肾阳方对该模型大鼠的疗效,并与较为公认可改善抑郁症的逍遥散进行疗效对比。结果:抑郁模型组大鼠Open-Field评分、糖水摄取量、5-HT、DA、NE明显低于空白组(P<0.05或P<0.01);各治疗组可改善大鼠Open-Field评分、糖水摄取量、5-HT、DA、NE,与模型组相比有统计学差异(P<0.05),各治疗组比较未见显著性差异。结论:温补肾阳方改善抑郁大鼠的抑郁状态与逍遥散相近,提示临床可采用温补肾阳法治疗抑郁症。

Objective ： To study the effect of prescription for warming and recupe - rating the kidney - yang on the behavior and monoamine neurotransmitters in the depression rat. Methods ：72 adult SD rats were randomly divided into a normal group, a depr - essive model group, the WBSYF with low dose group ,the WBSYF with middle dose group and the WBSYF with high dose group, the XYS with high dose group. Separate -d feeding,long -term unpredictable and middle stimulation stress were used for devel - opppppment of depression rat model. To oberseve the effect on depression rat mod- el treated by prescription for warming and recuperating the kidney - yang and compared with the Xiaoyao San which was accepted to treat depression. Regular determination of weight change in each group rats. Autonomic behavior was measured with Open Field test and concentration of amino neurotransmitter in hippoeampus （5 - HT, DA, NE ） were measured by ELISA. Results： The score of Open- Field,sucrose solution consumption,5 -HT,DA ,NE of depressive model group was obviously lower than that of control group （ P 〈 0.05 or P 〈 0.01 ） ; the scores of Open - Field, sucrose solution consump- tion,5 - HT,DA,NE of each treatment groups improved much better than those of model group（ P 〈 O. 05 ）. There is no significant difference among treatment groups （ P 〉 O. 05 ）. Conclusion ： The findings indicated that the efficacy of pre- scription for warming and recuperating the kidney - yang was similar to that of Xiaoyao San, so it could use prescription for warming and recuperating the kidney- yang to treat depression.
