胃癌中Krülppel样核转录因子-4的表达与核转录因子特异性蛋白-1和Notch跨膜受体-1表达的关系及意义 被引量:2

摘要 目的研究胃癌组织中Krülppel样核转录因子-4(KLF4)和核转录因子特异性蛋白-1(Sp1)、Notch跨膜受体-1(Notch1)的表达,探讨KLF4和Sp1、Notch1在胃癌组织中表达的相关关系及三者与胃癌临床病理的关系。方法采用免疫组织化学染色法,分别对76例胃癌手术标本及30名同期收集的正常胃组织标本中Sp1、Notch1和KLF4进行蛋白检测,分析三者的表达水平与临床病理的关系及KLF4的表达与Sp1、Notch1表达的关系。结果正常胃组织标本中可见KLF4蛋白强阳性表达率为77%,在胃癌组织中表达较弱或表达缺失;Sp1在胃癌组织中阳性表达率为63%,正常组13%;Notch1在胃癌组织中阳性表达为66%,明显高于正常胃组织的23%,且三者表达与胃癌分化程度、胃癌分期及淋巴结转移密切相关(P<0.05)。KLF4和Notch1在胃癌中表达呈负相关(r=-0.629,P<0.05),KLF4和Sp1在胃癌组织中表达呈负相关(r=-0.566,P<0.05)。结论 KLF4和Sp1、Notch1在胃癌组织中表达失衡可能对胃癌的发生发展起重要作用,三者可能成为胃癌预后的判断指标及潜在的治疗靶点。 Objective To investigate the expressions of KrLilppel-like nuclear transcription factor-4 (KLF4), specific protein-1 (Sp-1) and Notchl in gastric cancer, and to explore the correlations among KLF4, Spl and Notchl, and their relevance with the clinical pathology. Methods The expressions of Spl, Notchl and KLF4 in 76 specimens of gastric cancer and 30 of normal gastric tissue were detected with immunohistochemieal staining (S-P method). The correlations among KLF4, Spl and Notch1 as related to the clinical pathology of gastric cancer were then analyzed. Results The KLF4 showed strong positive expression (77%) in normal gastric tissue but was minimally or not detected in gastric cancer tissue. The expression of Spl was found in 63% of gastric cancer tissue vs 13% of normal gastric tissue. The positive rate of Notchl in gastric cancer tissue (66%) was significantly higher than that in normal gastric tissue (23%). In addition, these three factors were closely con'elated with differentiation, staging and lymph node metasta- sis of the gastric cancer (P〈0.05). The expression of KLF4 was negatively correlated with Notchl (r=-0.629, P〈O.05) and Spl (r=-0.566, P〈0.05) in gastric cancer tissue. Conclusion The disparity in expressions of KLF4, Spl and Notchl may play an important role in development and progression of gastric cancer, suggesting that these markers may be prognostic and potential as therapeutic targets for gastric cancer.
