芍药甘草汤防治支气管哮喘机理研究 被引量:13

摘要 目的:探讨芍药甘草汤防治支气管哮喘模型小鼠的作用机制。方法:采用卵蛋白(OVA)致敏剂制备小鼠哮喘模型,随机分正常对照组、模型对照组、地塞米松组、芍药甘草汤低、中、高剂量组6组,流式细胞术检测外周血中调节性T细胞CD4+CD25+百分比,观察肺组织形态学的改变,肺泡灌洗液(BLAF)中的细胞计数及分类。结果:模型组小鼠肺组织有明显炎性细胞浸润,渗出物增加,水肿明显,说明造模成功。芍药甘草汤中、高剂量组CD4+CD25+T细胞比例显著升高(P<0.05),低剂量组与模型对照组比较没有统计学意义。病理学组织观察示芍药甘草汤中剂量组、地塞米松组炎症性变化较芍药甘草汤高、低剂量组及模型组明显减轻。芍药甘草汤中、高剂量组与模型对照组相比较,细胞总数及EOS、Lym比例均降低(P<0.05)。芍药甘草汤低剂量组与模型对照组比较没有差异(P>0.05)。结论:芍药甘草汤可以抑制OVA致敏支气管哮喘小鼠各类炎性细胞的增生,调节肺组织中氧化/抗氧化系统的平衡,减轻氧自由基对肺组织结构的氧化损伤程度,调节哮喘小鼠体内CD4+CD25+T的优势表达,芍药甘草汤对支气管哮喘有一定的预防及治疗作用。 Objective : To investigate the mechanism of action of Decoction of Paeonia and Licorice on bronchial asthma in mouse. Methods : The asthma models of mice were made with ovalbumin (OVA) to sensitize. Then they were randomly divided into normal control group, model control group, dexamethasone group, treatment groups of low, medium and high dose. At last peripheral regulatory T cells CD4 ^+ and CD25 ^+ + percentage were determined with flow cytometry, morphological changes of lung tissue were observed, and the cell counting and classification in bronchoalveolar lavage fluid (BLAF) were also detected. Results: In medium and high dose groups the proportion of CD4 ^+ and CD25 ^+ T cells were significantly higher ( P 〈 0. 05 ) low-dose group compared with the model control group. Pathological tissue changes , but was not statistically significant in observation showed the inflammatory in medium dose group and dexamethasone group were significantly reduced than in high and low dose groups and model control group. The total cellular score, EOS and Lym ratios were cut down in medium and high dose groups compared with the control group (P 〈 0. 05 ), but no statistical significant difference showed between low dose group and model control group (P 〉 0. 05 ). Conclusion : Decoction of Paeonia and Licorice can inhibit various inflammatory cellular proliferations in OVA-sensitized asthmatic mice, regulate the balance of oxidant / antioxidant system in lung tissue, reduce the degree of oxidative damage caused by oxygen free radicals on lung tissue structure, and adjust the advantage expressions of CD4 ^+ and CD25 ^+ T cells in asthma mice. Decoction of Paeonia and Licorice has some therapeutic effect on the prevention and treatment of bronchial asthma
