阳离子脂质体基因转染研究

Study on Gene Transfection Mediated by Cationic Liposomes

摘要 目的:评价阳离子脂质体介导基因转染宫颈癌细胞的转染效果,优化对宫颈癌细胞的转染条件。方法:以荧光素酶基因pGL3为报告基因,分析影响阳离子脂质体Lipofectamine 2000转染Hela效率的因素。结果:通过细胞转染实验,得出脂质体与pDNA的稀释液混合较佳等待时间为30 min,脂质体/pDNA复合物与细胞接触的转染时间为4 h,转染效率较高。结论:Lipofectamine2000 Hela细胞转染效率较高。 Objective: To evaluate the gene transfection efficiency of cationic liposome to cervical carcinoma cell,in addition,to optimize the transfection conditions.Method: the transfection efficiency of liposome were examined using Luceferase as reporter gene.Result: Lipofectamine 2000 could get higher transfection efficiency when dilutin time was 30 min before complexing and the optimum transfection time was 4 h.Conclusion: Lipofectamine 2000 obtained higher transfectin efficiency in Hela cells.
