摘要
[目的]通过在卷叶贝母液体培养基中添加不同氨基酸研究其对卷叶贝母培养物中有效组分的影响。[方法]分别在卷叶贝母液体培养基中添加不同浓度的苯丙氨酸、酪氨酸和色氨酸,并利用分光光度法测定培养25 d后其培养物中总生物碱含量。[结果]添加苯丙氨酸、酪氨酸和色氨酸前体物均能显著地提高卷叶贝母培养物的增殖倍数和总生物碱含量。[结论]添加浓度为0.50 mmol/L的酪氨酸试验组其培养物增殖倍数最高为2.60,以添加浓度为0.50 mmol/L的色氨酸试验组其总生物碱含最高,为0.212%。
[Objective] The aim was to study the effect of different amino acid on the active ingredient of Fritillaria cirrhosa D.Don culture.[Method] The Fritillaria cirrhosa D.Don was cultured in the liquid medium that contained different concentration of phenylalanine,tyrosine and tryptophan.After 25 days incubation,the total alkaloid of Fritillaria cirrhosa D.Don culture was determined by spectrophotometry.[Result] The results indicated that the total alkaloid and the proliferation times of the Fritillaria cirrhosa D.Don culture were significantly improved.[Conclusion] The proliferation times of the culture obtained from the group that contained 0.50 mmol/L tyrosine could reach up to 2.60 and the total alkaloid of Fritillaria cirrhosa D.Don of the culture obtained from the group which contained 0.50 mmol/L tryptophan could reach up to 0.212%.
