摘要 [目的]建立扛板归中大黄素的提取和含量测定方法。[方法]采用反相高效液相色谱法测定扛板归中大黄素的含量。色谱条件:色谱柱为Hypersil C18,流动相为甲醇-水-冰醋酸(V/V/,V 87∶13∶215),流速为110 m l/m in,检测波长为254 nm,柱温为室温。[结果]在1~40μg/m l范围内,大黄素呈良好的线性关系,线性回归方程为Y=112.89X-8.912(r=0.999 9)。扛板归中的大黄素提取最优工艺条件为乙醇的浓度为80%,乙醇的用量为35 m l,提取时间为2 h。平均加样回收率为99.16%(RSD=1.49%)。[结论]该方法简便、快速、准确,可用于扛板归中大黄素的提取与含量测定。 [Objective] To extract and determine the content of emodin in Polygonum perfoliatum L.[Method] The content of emodin was determined by RP-HPLC.A column of Kromasil C18 was used and a mixture of methanol-water-glacial acetic acid(87∶13∶215) was used as the mobile phase at a flow rate of 110 ml/min.The UV detection wavelength was 254 nm and the column temperature was room temperature.[Result] The emodin showed a good linear relationship at the range of 1-40 μg/ml,and the linear regression equation was Y=112.89X-8.912(r=0.999 9).The optimum conditions for the extraction of emodin from Polygonum perfoliatum were: 80% ethanol concentration,35 ml ethanol dose and 2h extraction time.The average recovery rate was 99.16%(RSD=1.49%).[Conclusion] The method was simple,rapid and accurate.It could be used for the determination of emodin in Polygonum perfoliatum L..
