摘要
[目的]研究杜仲总黄酮对新生大鼠颅盖骨体外培养成骨细胞护骨素的mRNA及蛋白表达的影响。[方法]分别加入浓度为10、100、200μg/ml的杜仲黄酮到SD大鼠颅骨分离培养的成骨细胞中,用Realtime-PCR法检测杜仲总黄酮对护骨素的mRNA表达的影响;采用细胞免疫荧光技术检测杜仲总黄酮对护骨素蛋白表达的影响。[结果]与对照组比较,杜仲总黄酮能明显促进成骨细胞护骨素的mRNA和蛋白质的表达。[结论]杜仲总黄酮成分能直接在RNA和蛋白质水平促进体外成骨细胞中护骨素的表达。
[Objective] To research the effect of total flavonoids of CORTEX EUCOMMIAE on the expression of osteoprotegerin mRNA and protein by osteoplast in vitro cultured from the cranium of newly-born rat.[Method] 10,100,200 μg/ml total flavonoids of CORTEX EUCOMMIAE was respectively added into the osteoplast isolated from the cranium of SD rats,and then the impact of total flavonoids of CORTEX EUCOMMIAE on mRNA expression of osteoprotegin was detected by Realtime-PCR method,and the impact on protein expression was detected by immunofluorescence.[Result] Compared with the control group,the total flavonoids of CORTEX EUCOMMIAE could significantly promote the mRNA and protein expression of osteoprotegin by osteoplast.[Conclusion] The total flavonoids of CORTEX EUCOMMIAE could directly promote the expression of osteoprotegin by osteoplast at the RNA and protein level.
