重组人proEMAP Ⅱ/p43抑制新生血管生成与细胞凋亡关系研究 被引量:1

摘要 目的研究p43蛋白抑制新生血管生成是否通过诱导细胞凋亡来实现。方法用不同浓度的p43蛋白(0,10,50,100μg/ml)作用于HUVEC细胞12 h,检测细胞体外管腔形成能力的变化,同时检测细胞周期变化,并通过胞内总半肽天冬酶(caspase)活性检测、caspase3和caspase7基因水平的变化及DNA片段化检测等方法,研究p43蛋白对HUVEC细胞凋亡的影响。结果与结论 p43蛋白可明显抑制HUVEC细胞体外管腔形成,并随着p43蛋白浓度的升高抑制作用不断增强;不同浓度的p43蛋白对HUVEC细胞周期没有明显影响,不能诱导HUVEC细胞凋亡。p43蛋白抑制新生血管的生成不是通过诱导细胞凋亡来实现的。 Objective To study whether p43 protein's inhibits angiogenesis can be achieved by inducing apoptosis. Methods With different concentrations of p43 protein in human umbilical vein endothelial cells ( HUVEC ) for 12 hours, the change of tube formation, cell cycle assay, and total easpase activity assay were detected. The mRNA of caspase3, caspase7 assay and DNA ladder assay were used to study p43 protein on HUVEC apoptosis. Results and Conclusion p43 Protein could inhibit tube formation of HUVEC in vitro and the increase in p43 protein concentration can enhance the inhi- bition of tube formation. Difference in the concentration of p43 protein has no effect on HUVEC cell cycle, unable to induce apoptosis of HUVEC. 1943 Protein does not inhibits angiogenesis via inducing apoptosis.
