表皮生长因子受体在DNA损伤修复中的作用机制新进展

摘要 表皮生长因子受体(epidermal growth factor receptor,EGFR)在许多人类恶性肿瘤中存在过表达或突变,近期大量的研究表明EGFR和/或其下游成分可以进入细胞核,在核内发挥转录调节和信号转导功能,其中一个重要的功能是促进DNA双链断裂(double strands breaks,DSBs)的修复.EGFR主要通过核移位及PI3K-Akt通路、Ras-Raf-MAPK通路与DNA修复的关键成分如DNA-PK,ATM,Rad51和BRCA1等作用促进DSBs的修复.放化疗诱导的肿瘤细胞DNA损伤后的修复目前正成为一个越来越有吸引力的靶点,靶向EGFR或其下游信号抑制DNA损伤修复的治疗具有重要的临床意义和应用前景.
