20份葡萄种质亲缘关系的SSR分析

Analysis of genetic relationship among 20 Vitis germplasm resources by SSR markers

利用SSR标记对20份葡萄材料进行了基因组多态性分析,从245对引物中筛选出19对用于葡萄的SSR扩增。共扩增出144条带,均为多态性条带,多态性百分率为100%。根据SSR扩增结果,利用NTSYSpc2.10e软件进行Jaccard相似性系数分析,20份葡萄材料的遗传相似系数为0.630～0.900,平均遗传相似系数为0.765。通过UPG-MA进行聚类分析,在遗传相似系数0.665处,20份葡萄材料可分为4大类群。第1类包含欧洲葡萄、美洲种群的种间杂种和欧美杂种;第2类包含我国南方野生资源刺葡萄、毛葡萄、复叶葡萄、菱状叶葡萄、蘡薁葡萄和华东葡萄;第3类为山欧杂种;第4类为山葡萄。山葡萄与欧亚种、美洲杂种的亲缘关系较远,欧亚种与美洲种之间亲缘关系较近。

Twenty Vitis germplasm resources were used as materials for analyzing their genome polymorphism by SSR markers in the present study.Nineteen pairs of polymorphic primers selected from 245 pairs were used for SSR amplification.144 DNA fragments were generated and all of them were polymorphic bands（the percentage of polymorphic band,PPB=100%）.Jaccard similarity coefficients of the SSR amplification results were analyzed by NTSYSpc2.10e software and the genetic similarity coefficient was 0.630 to 0.900 with the average genetic similarity coefficient of 0.765.The clustering dendrogram constructed by UPGMA method indicated that 20 Vitis materials could be divided into four major groups at the similarity coefficient of 0.665.The first group included V.vinifera,American grape hybrids and Euro-American hybrids.The second group included V.davidii Foёx.V.quinquangularis Rehd.,V.piasezkii Maxim.,V.hancockii Hance,V.thunbergii Sieb..Zucc.and V.pseudoreticulata W.T.Wang.The third group included the hybrids（V.amurensis×V.vinifera） ×V.amurensis.The fourth group included V.amurensis Rupr.materials.The results suggested that the genetic correlations were much lower between V.amuensis and V.vinifera,or Euro-American hybrid,but higher between V.vinifera and Euro-American hybrid,or American hybrids.
