重症急性胰腺炎患者血清差异蛋白质的表达

摘要 目的:用蛋白质组学技术比较重症急性胰腺炎(SAP)与轻症急性胰腺炎(MAP)患者血清的差异蛋白,探讨SAP特异蛋白的表达。方法:血清标本取自10例急性胰腺炎(AP)患者,其中5例SAP,5例MAP,用双向凝胶电泳检测两类患者的蛋白表达,并将这些差异表达蛋白点进行质谱鉴定,Mascot数据库检索。结果:比较双向电泳图谱发现15个差异表达蛋白点,经质谱分析,成功鉴定出10种蛋白质,在SAP患者其中4种表达上调,6种表达下调。数据库查询结果,4个表达上调蛋白分别为补体4A、结合珠蛋白亚型2、结合珠蛋白和载脂蛋白L1;6个表达下调蛋白分别为四连接素、谷胱甘肽过氧化物酶3、丝氨酸蛋白酶抑制剂亚型F1、丛生蛋白亚型-1、转甲状腺素蛋白、丝氨酸蛋白酶抑制剂A1亚型2。结论:SAP患者血清有差异蛋白质表达,这可能和SAP发病机制相关。 Objective: To compare the serum protein differences between patients with severe acute pancreatitis (SAP) and mild acute pancreatitis (MAP) with proteomic analysis in order to explore the differentially expressed proteins in SAP patients. Methods: Serum samples from 5 patients with SAP and 5 with MAP were collected. Two-dimensional electrophoresis (2-DE) was applied to examine the protein expression in the samples. The spots of differentially expressed proteins from the two groups were further identified by mass spectrometry. The data were searched in the Mascot database. Results: In SAP patients, 15 spots of differentially expressed proteins were detected by 2-DE, from which 10 kinds of proteins were successfully identified by mass spectrometry. Searching of Mascot database revealed 4 up-regulated proteins [ complement-4A, haptoglobin isoform 2, Haptoglobin and apolipoprotein-L1 ( ApoL1 ) I and 6 down-regulated proteins[ tetranectin, glutathion peroxidase- 3 ( GPX-3 ), serpin-F1 pigment, clusterin isoform-1, transthyretin, and Serpin-A1 isoform-2 ]. Conclusion: Differentially expressed proteins were detected from the sera of patients with SAP, which might be related to the pathogenesis of SAP.
