慢性肾功能衰竭大鼠肾皮质基因组DNA表观遗传学变化的MSAP分析 被引量:3

Detection of DNA Methylation Changes in Chronic Renal Failure Rats Using Methylation-sensitive Amplification Polymorphism(MSAP)

摘要 以灌胃腺嘌呤诱导的慢性肾功能衰竭Wistar大鼠为实验材料,用高效液相色谱法(HPLC)检测了大鼠血清Hcy浓度的变化;应用甲基化敏感扩增多态性(methylation-sensitive amplification polymorphism,MSAP)技术,检测了慢性肾衰大鼠肾皮质基因组DNA甲基化程度的变化,并对MSAP差异片段进行了分析.结果发现:(1)慢性肾衰大鼠血清Hcy浓度由正常的(10.56±1.20)μmol/L上升为(16.64±1.57)μmol/L,P<0.05,差异具有显著性.(2)慢性肾衰大鼠肾皮质基因组总甲基化率由正常的(41.95±1.94)%降低为(38.43±3.27)%,P<0.05,差异具有显著性.(3)甲基化差异片段涉及多个基因,其中包括多个甲基化转移酶相关基因.结果表明慢性肾功能衰竭大鼠会出现高同型半胱氨酸血症和肾皮质基因组总甲基化水平的降低,多个基因内部或上下游DNA序列出现甲基化状态变化. To investigate whether DNA hypomethylation is present in rats with chronic renal fairlure and to assess the possible sequence fragments related to the DNA hypomethylation, adenine-induced Chronic Renal Failure(CRF) rat as animal model was constructed and detected the change of Hcy concentration in the serum of rats by the method of HPLC.The extent and pattern of methylation in the genome from the cortex of kidney of CRF rats were analyzed by the technique of methylation-sensitive amplified polymorphism(MSAP).The results showed that:(1) Concentration of Hcy in the serum of CRF rats raised up to(16.64±1.57)μmol/L,compared to(10.56±1.20)μmol/L under normal conditions,with P0.05 showing significant difference.(2) Methylation rate in the genome from the cortex of kidney of CRF rats reduced to(38.43±3.27)%,compared to(41.95±1.94)%under normal conditions, with P0.05 showing significant difference.(3) Differentially-methylated fragments involved many genes including several genes related to methyltransferase.The results suggest CRF rats suffer from hyperhomocysteinemia and the pattern of methylation in the genome changes significantly.
