摘要
目的:探讨三种射干提取物止咳作用的强弱及其药效动力学过程。方法:采用一次性灌胃给药,以浓氨水雾化诱咳的方法,观察记录小鼠的咳嗽潜伏期和2分钟内咳嗽次数,建立相应的时间-效应方程及剂量-效应方程,计算相关药效动力学参数。结果:三种射干提取物在小鼠体内均呈一室模型特征,2号、6号、7号止咳效应半衰期分别为1.314h、2.149 h、0.538 h,效应消除半衰期分别为8.226h、3.971 h、8.026 h,药效作用期分别为5.73h、7.74 h、7.38 h,效应达峰时间分别为为1.0h、2.0h、2.0h;2号、6号、7号延长咳嗽潜伏期效应半衰期分别为0.272h、8.427h、0.553h;效应消除半衰期分别为2.579h、8.428h、5.259h;药效作用期分别为4.14h、6.64 h、7.29 h,效应达峰时间分别为1.0h、1.0h、2.0h;2号、6号、7号延长咳嗽潜伏期最低起效剂量分别为24.83、1.02、2.12mg/kg,抑制咳嗽次数的最低起效剂量分别为5.06、0.34、0.41mg/kg。结论:三种射干提取物均具有明显的止咳作用,并且起效剂量低、起效迅速、药效作用时间持久。
Objective:To study pharmacodynamics of the three belamcanda chinensis extracts and effect on hustenmildernd.Method: The purpose of this study was to describe-following the intragastric administration once and cough induced by ammonia-observing the cough latent period and recording times during two minutes,some correlations between time and effect,dose and effect,as well as how these variables fit in a model.Results:We found that three belamcanda chinensis extracts serve as one compartment model in mice,number two,six and seven,hustenmildernd effected half life are 1.314h,2.149 h,0.538 h,effect eliminated half life are 8.226h,3.971 h,8.026 h,efficacy time are 5.73h,7.74 h,7.38 h,effect peak time are 1.0h,2.0h,2.0h respectively,we also found that number two,six and seven of belamcanda chinensis extracts,prolong cough latent period effected half life of are 0.272h,8.427h,0.553h,effect eliminated half life are 2.579h、8.428h、5.259h,efficacy time are 4.14h,6.64 h,7.29 h,effect peak time are 1.0h,2.0h,2.0h respectively,number two,six,seven of belamcanda chinensis extracts,prolonged cough onset latency of the lowest dose are 24.83,1.02,2.12mg/kg,the minimum inhibitory dose onset on cough times are 5.06,0.34,0.41mg/kg.Conclusion: Three belamcanda chinensis extracts have obvious hustenmildernd effect and characteristics of low-dose,rapid and long-lasting efficacy.
