摘要
目的探讨不同地理株白纹伊蚊(Aedes albopictus)线粒体细胞色素氧化酶亚基Ⅰ(mtDNA-COI)基因序列特征、种群差异和分化程度。方法扩增不同地理株白纹伊蚊mtDNA-COI,并从网上下载部分白纹伊蚊mtDNA-COI进行序列测定、分析和比对。结果①获得mtDNA-COI长度为415 bp,碱基A+T含量为70.3%,颠换率64.7%。②个体存在6种单倍型有41个,单倍型家系网络显示所有的单倍型都由一种单倍型进化而来。分子变异等级分析(AMOVA)结果显示,该遗传分化主要来自种群内(71.9%)。遗传距离结果显示不同地理种群出现较小程度的分化(Fst=0.28136),可能由遗传漂变引起。结论不同地理株白纹伊蚊出现轻度的遗传分化,很可能由遗传漂变引起,主要由种群内变异引起。
Objective To study the characteristics of mitochondrial DNA cytochrome oxidase sunbunit I (mtDNA-COI) gene and the population genetic variation and the diversity level in different geographic strains of Aedes albopictus. Methods Downloaded from Genbank, the amplified mtDNA-COI gene sequences in different geographic strains Aedes albopictus were sequenced, analyzed and compared with the others in the Genbank. Results ① The COI fragment contained 415 nucleotides, with 8 polymorphic sites. The A+T content of COI gene was 70.3%. The rate of transversion was 64.7 %. ② Six hapiotypes were obtained 41 specimen. Genealogical relationship and minimum spanning trees indicated the evolutionary relationship among haplotypes. AMOVA analysis showed that the genetic difference mainly came from variation within population whose the percentage was 71.9% and the degree of that was lower (Fst= 0. 28136). Conclusion The genetic difference existed in different geographic strains of Aedes albopictus , but the degree was lower. This difference mainly came from variation within the population and might caused by genetic drift.
