鸭圆环病毒ORF3基因的克隆及序列分析 被引量:4

摘要 为了解鸭圆环病毒(DuCV)不同分离株ORF3基因变异情况及分析其同源性,本研究从临床病料中分离10株DuCV,扩增ORF3基因并测定其序列。通过与已知序列比对分析发现,DuCV ORF3基因分为两种基因型,大小分别为237 bp和297 bp。ORF3基因同一基因型之间的核苷酸同源性为95.8%~100%,氨基酸同源性为88.6%~100%,而不同基因型之间的核苷酸同源性仅为87.8%~91.6%,氨基酸同源性仅为72.2%~81.0%。 To elucidate the ORF3 gene variations of different duck circovirus(DuCV) isolates,the ORF3 gene sequences of 10 DuCV isolates,isolated from the clinical tissue samples,were analyzed and compared with that of reference DuCV strains.Sequencing results indicated that the ORF3 gene of the isolates had 2 genotypes which were 237 nt and 297 nt,respectively.The nucleotide homology of ORF3 gene in the same genotype was between 95.8% to 100%,but it was only 87.8% to 91.6% between the different genotypes.Furthermore,the deduced amino acid homology of this gene in the same genotype was between 88.6% to 100% and it was only 77.2% to 81.0% between the different genotypes.
