摘要 目的:探讨体外应用表没食子儿茶素没食子酸酯(EGCG)对人乳腺癌MCF-7细胞增殖、侵袭及其对血管内皮细胞生长因子-C(VEGF-C)表达的影响。方法:体外培养MCF-7细胞,设立对照组及ECa2G实验组,应用MTT法及Transwell侵袭实验观察不同浓度ECA2G对MCF-7细胞增殖及侵袭的影响;应用免疫细胞化学显色及免疫印迹法观测EGC/3对VEGF-C分泌及蛋白表达水平的影响。结果:一定浓度的EGCG可抑制人乳腺癌MCF-7细胞的增殖及侵袭,同时可抑制VEGF-C的分泌及蛋白的表达。结论:ECA2G可以通过抑制肿瘤细胞的增殖、侵袭及VEGF-C的表达来发挥抗肿瘤的作用。 Objective: Effects of epigallocateclin-3-gallate (EGCG) were examined on human breast cancer cell line MCF-7 in vitro for the proliferation, invasion and expression of vascular endothelial growth factor-C (VEGF-C). Methods: In vitro, human breast cancer cell line MCF-7 was treated with ECK2G at different concentrations, and then the proliferation and migration of cells were assayed by MTT method and Transwell respectively. Immunocytochemistry and Western blot assay were used to investigate the secretion and expression of VEGF-C respectively. Results: ECK2G treatment inhibited the proliferation and migration of MCF-7 cell, and it also reduced the secretion and protein expression of VEGF-C. Conclusion: EGCG can exert the anti-tumor effect by inhibiting the proliferation and migration of tumor cells, as well as the secretion and protein expression of VEGF-C.
