人参皂苷Rh2调节白血病细胞增殖分化的作用 被引量:5

摘要 目的:观察人参皂苷Rh2对白血病细胞HL60增殖分化的影响。方法:免疫印迹法及流式细胞分析法分别检测未处理组HL60细胞及药物处理组HL60细胞的核增殖抗原(PCNA)及成熟粒细胞表面抗原分子CD15的表达水平。结果:药物处理组的HL60细胞体积增大,形状不规则;与未处理组HL60细胞相比,药物处理组的HL60细胞增殖速度明显减慢,PCNA的表达水平降低;未处理组HL60细胞表达CD15的阳性率为42.22%,药物处理组HL60细胞表达CD15的阳性率为73.31%,表达水平明显升高。结论:人参皂苷Rh2对白血病细胞HL60,具有抑制增殖促进分化的作用。 Objective: To evaluate the effects of ginsenoside-Rh2 on the proliferation and differentiation of HL60 cells. Methods: Before and after being treated with ginsenoside-Rh2, the growth of the HL-60 cells was observed under an inverted phase contrast microscope. The proliferating ceil nuclear antigen (PCNA) was assessed by Western blotting and mature myeloid differentiation marker CD15 was quantified by flow cytometry. Results: The experimental group of HL60 cells were significantly enlarged in size and weakened in proliferation compared to wild type HL60 cells (control). The level of PCNA was downregulated, meanwhile CD15 was upregulated. Conclusion: Ginsenoside-Rh2 can inhibit the growth and induce the differentiation of HL60 cells.
