T淋巴细胞侵袭转移诱导蛋白1在小鼠子宫内膜基质细胞-胚泡共培养体系中对胚泡黏附的影响

摘要 目的:检测不同浓度T淋巴细胞侵袭转移诱导蛋白1(Tiam1)抗体下,小鼠着床窗子宫内膜基质细胞内的Tiam1蛋白表达及其对基质细胞-胚泡共培养体系中胚泡黏附的影响,探讨Tiara1对小鼠胚胎着床的影响。方法:提取着床窗小鼠子宫内膜基质细胞并构建基质细胞-胚泡共培养体系;分别用免疫细胞化学和免疫印迹法检测不同浓度抗Tiam1抗体时着床窗基质细胞Tiam1蛋白的表达及其对胚泡黏附的影响。结果:抗体干预后着床窗基质细胞内的Tiam1蛋白表达水平降低;在不同抗体浓度下胚泡黏附率有差异。结论:随Tiam1抗体浓度增加,着床窗小鼠子宫内膜基质细胞Tiam1蛋白表达量及胚泡黏附率降低。表明Tiam1可能在胚泡植入过程起重要的作用。 Objective: To observe the expression of T-lymphoma invasion and metastasis inducing protein 1 (Tiaml) on endometrial stromal cell and its influence on blastocyst adhesion in mice endometrial stromal cells-blastocysts co-culture system. Methods: The endometrium stromal cells were separated from mice to establish endometrial stromal cells-blastocysts co-culture system in vitro. The rate of blastocyst adhesion was recorded. The expression of Tiara1 at protein level with the different antibodies concentrations was evaluated by immunohistochemistry and Western blotting. Results: The levels of Tiaml protein and the blastocyst adhesion rate decreased in mice endometrial stromal cells during the implantation window under anti-Tiaml antibody intervention, with a statistical significance in each group (P〈0.05). Conclusion: With the increasing concentration of anti-Tiaml antibody, the number of blastocyst adhesion and the protein expression is reducing. All the results indicate that Tiaml may be involved in embryo implantation and may play an important role in the blasto- cyst adhesion.
