摘要
目的:探讨压力作用不同时间对人牙周膜成纤维细胞表达TGF-β1的影响。方法:本实验采用细胞加载装置,对人牙周膜成纤维细胞施加1.0MPa的压力,分别持续10min、30min、50min,通过酶联免疫吸附实验,分别检测细胞受力后6h、12h、18h、24h时TGF-β1的含量。结果:加载10min组在细胞受力后的各个时段TGF-β1含量与对照组相比无显著性差异(P>0.05);加载30min组(0.51±0.06)和加载50min组(0.46±0.06)TGF-β1含量在细胞受力后的第12h同对照组(0.78±0.08)相比明显降低(P<0.05)。结论:在一定加载时间内,人牙周膜成纤维细胞合成TGF-β1量随着压力作用时间的延长而有所降低。
Objective: To investigate the relationship of transforming growth factor-β1(TGF-β1) secreted by human periodontal ligament fibroblasts(HPLF) to duration times of pressure.Methods: We treated cells for 10min,30min and 50min respectively with 1.0MPa pressure by a cellular mechanical research system.TGF-β1 in the supernant was measured at 6h,12h,18h and 24h after stimulation by enzyme link immunosorbent assay(ELISA).Results: The results showed that cells treated for 10min had no difference in TGF-β1 content compared with control group at different duration times.TGF-β1 of 30min(0.51±0.06) and 50min(0.46±0.06) groups was lower than that of control group(0.78±0.08) at 12h after stimulation respectively.Conclusion: In certain range of duration times of pressure,mechanical pressure can suppress TGF-β1 synthesis by HPLF in a duration-time dependent manner.
