龙葵多糖对红细胞CR1数量及S_(180) A和H_(22)荷瘤小鼠机体红细胞免疫力的影响 被引量:5

Enhancement of Erythrocyte Immunity in S_(180) OR H_(22)-bearing Mice by Solanum Nigrum Polysaccharide Through Increasing Quantity and Activity of Red Cell CR1

摘要 目的研究龙葵多糖对S180及H22荷瘤小鼠红细胞补体受体1(CR1)数量及活性的影响,探讨龙葵多糖抗肿瘤作用的红细胞免疫机制。方法采用体内移植S180和H22肿瘤细胞株造模的方法,评价龙葵多糖的体内抗肿瘤疗效;红细胞免疫花环实验检测红细胞免疫黏附肿瘤细胞的能力;CR1单克隆抗体标记,流式细胞术检测荷瘤小鼠红细胞CR1数量;酵母多糖免疫黏附荷瘤小鼠红细胞的能力评价荷瘤小鼠红细胞CR1受体的活性;比色法测定荷瘤小鼠红细胞膜唾液酸含量。结果龙葵多糖能够抑制S180荷瘤小鼠实体瘤生长,延长H22荷瘤小鼠生存时间;提高荷瘤小鼠红细胞免疫花环率(DTER);增加荷瘤小鼠红细胞CR1受体的数量和活性;增加荷瘤小鼠红细胞膜唾液酸含量。结论龙葵多糖通过提高荷瘤小鼠红细胞膜上唾液酸含量,进而提高CR1的活性和数量,改善和提高荷瘤小鼠红细胞免疫黏附能力,发挥抗肿瘤的作用。 OBJECTIVE To study the effect of solanum nigrum polysaccharide(SNP) on the quantity and activity of red cell CR1 receptor in S180 or H22-bearing mice.METHODS The antineoplastic activity of SNP were tested in vivo using transplantable S180 and H22 tumors;erythrocytic immunity was assayed by red blood cell immune complexes rosette(RBC-ICR).CR1 quantity was tested by flow cytometer;erythrocytic immunity activity was assayed by red blood cell immune complexes rosette(RBC-ICR);sialic acid content of erythrocyte membrane was tested by colorimetry assay.RESULTS SNP could inhibit the tumor weight of S180-bearing mice and prolong the survival time of H22-bearing mice.SNP could increase the DTER,quantity or activity of CR1 receptor,and sialic acid content.CONCLUSION The anti-tumor effect of SNP in tumor-bearing mice is due to its ability to improve the sialic acid content,quantity and activity of CR1 acceptor of the red cell membrane and to enhance the immunity of the red blood cell.
