电刺激硬脑膜对三叉神经节连接蛋白40表达和痛阈的影响 被引量:1

摘要 目的:探讨三叉神经节中连接蛋白40在偏头痛发作中的临床意义。方法:将健康成年SD大鼠随机分为假手术组、对照组、实验0h组、实验12h组、实验24h组、实验48h组,采用电刺激大鼠上矢状窦区硬脑膜模型。应用免疫荧光和Westem blot技术观察各组三叉神经节连接蛋白40表达,应用von frey纤维丝测定大鼠眼周皮肤的痛觉阈值。结果:免疫荧光显示实验0h组、实验12h组、实验24h组连接蛋白40的表达均高于假手术组、对照组和实验48h组,于1 2h达表达高峰(P<0.05)。各组左右侧无统计学差异(P>0.05)。Western blot显示实验0h组、实验12h组、实验24h组连接蛋白40的表达均高于假手术组、对照组及实验48h组(P<0.05)。各组左右侧无统计学差异(P>0.05),于12h达表达高峰。Von Frey纤维丝测定显示实验组大鼠实验前1h、实验刺激后2h、12h、24h、48h左、右眼周皮肤的痛觉阈值呈先下降后上升的趋势,于12h达低点,48h基本恢复正常。结论:电刺激大鼠上矢状窦区硬脑膜可上调三叉神经节连接蛋白40的表达,降低大鼠面部的痛觉阈值,增强面部的痛觉敏化,表明连接蛋白40在偏头痛的发生及发展中可能起到一定作用。 Objective:To investigate the role of connexin 40 of trigeminal ganglion in migraine attack. Methods:Healthy adult SD rats were randomly divided into sham surgery group,control group,experimental group which according to different investigate time following the stimulation of rat dual matter adjacent to the superior sagittal sinus to divided into four subgroups.The changes in expression of connexin 40 of each group using immunofluorescence technique and Western blot analysis were observed. The pain threshold of periorbital skin using von Frey hair to monitor was also evaluated.Results: Immunofluorescence showed that the connexin 40 expression of experimental 0 h group,experimental 12 h group,experimental 24 h group were extremely higher than that of sham surgery group,control group and experimental 48 h group(P0.05).The top time point was at 12 h.In each group of both sides the expression was also equal(P0.05).Western blot analysis showed that the connexin 40 expression of experimental 0 h group,experimental 12 h group and experimental 24 h group much intensity higher than that of sham surgery group,control group and experimental 48h group(P0.05).The top time point was 12 h.In each group the expression was equal at both sides.Von Frey hairs study showed that the periorbital skin pain threshold of minus before 1 h group,the 2 h group,12h group,24h group and 48h group decreased at first and than increasd,the lowest timepoint at 12 h group and returned to normal level at 48 h.Conclusion:Following the stimulation of rat dual matter adjacent to the superior sagittal sinus expression of connexin 40 increased and pain threshold of periorbital skin.That connexin 40 may contribute to migraine attacks.
