摘要 目的研究驱虫斑鸠菊的血清药物化学。方法通过比较驱虫斑鸠菊提取物、给药及未给药大鼠血清的HPLC指纹图谱,确定灌服驱虫斑鸠菊后大鼠血中移行成分。结果经多次重复试验后,确定血清的处理方法为每100μL大鼠血清加入1 mL乙腈沉淀蛋白。驱虫斑鸠菊提取物给药组大鼠血清的HPLC图谱与不给药组空白血清对比有明显不同。含药血清中出现12个移行成分,其中有6个成分含量较高,且与提取物HPLC图谱在相同保留时间出峰,可能为原形成分,4种成分可能为代谢物。结论本研究确定了驱虫斑鸠菊的血中移行成分,为阐明驱虫斑鸠菊治疗白癜风药效物质成分奠定了基础。 Objective To investigate the serum pharmacochemistry of Vernohia anthelmintica.Methods HPLC-UV chromatogram was compared among the peaks of Vernonia anthelmintica extracts,serum with drug and blank serum to determine the transitional blood components in rats after the administration of extracts.Results After repeated experiments,we determined the better method to deal with the serum.We added 1 mL ethyl cyanide into 100 μL rat serum to precipitate the proteins.Twelve compounds were obtained,6 of which were found with high content and had the same retention time with those in the HPLC chromatogram of extracts,and another 4 compounds might be metabolites.Conclusion The compounds are absorbed into the blood and their metabolites are the effective constituents.The original crude drugs are the main ingredients in Vernonia anthelmintica.
