摘要
目的对膀胱癌腔内种植与膀胱黏膜微环境变化的关系进行初步研究。方法采用胰蛋白酶灌注膀胱的方法对Balb/c雌小鼠的膀胱黏膜进行损伤处理,并对损伤及未损伤组膀胱进行病理检查;ELISA法检测膀胱黏膜损伤与未损伤时小鼠膀胱CXCL12(SDF-1)含量的变化。通过观察Balb/c、nu/nu裸小鼠膀胱灌注肿瘤细胞后成瘤率对膀胱黏膜损伤与种植转移的关系进行评价。结果经0.25%胰蛋白酶损伤后饲养24h的小鼠的膀胱出现明显充血、水肿和炎症细胞浸润;胰蛋白酶损伤后的膀胱CXCL12(SDF-1)含量明显增加(P<0.05)。出现膀胱黏膜损伤和炎症的裸小鼠膀胱腔内种植的发生率更高。结论膀胱黏膜损伤、炎症及趋化因子CXCL12(SDF-1)的高表达与膀胱癌腔内种植关系密切。
Objective To study the relationships between bladder carcinoma cells implantation and bladder microenvironment in mouse' bladder.Methods The mucosa of urinary bladder was damaged by trypsin,then the damaged bladder were pathology tested,and CXCL12(SDF-1)expression in the tissue were also examed.The offspring cell lines EJ-m3,with high invasive ability were instilled into the bladder of athymic mouse which were damaged by trypsin,to appraise the relationships of bladder carcinoma cells implantation with the mucosa of urinary bladder damage.Results The mucosa of urinary damaged bladder by trypsin was engorgement,dropsy and inflammatory cells infiltrating.The expression of CXCL12(SDF-1) of mouse bladder which were damaged by trypsin was higher than those of controls(P0.05),and the neoplasm formation rate of athymic mouse in damaged groups by trypsin was higher than control group(P=0.004).Conclusions The engorgement,dropsy and inflammatory cells infiltrate,and the expression of CXCL12(SDF-1) of mouse' bladder have significant relationships with bladder carcinoma cells implantation in bladder.
