摘要
目的吗啡持续刺激对C57 BL/6小鼠骨髓来源的树突状细胞成熟的影响。方法体外培养C57 BL/6小鼠骨髓来源的树突状细胞,培养过程中给予吗啡刺激,观察细胞形态的变化;应用酶联免疫吸附法检测树突状细胞分泌细胞因子IL-12水平的变化。结果吗啡干预各组树突状细胞中细胞因子IL-12分泌水平较对照组显著升高。结论吗啡持续刺激能够促进小鼠树突状细胞的成熟。
Objective To investigate the effects of sustained morphine stimulation on the bone marrow-derived dendritic cell(DC) maturation.Methods The primary C57BL/6 mouse bone marrow-derived dendritic cells(4-6weeks) were cultured in vitro.In course of cell culture morphine was added at the same time.The cell supernatants were collected at 7 days and interleukin-12(IL-12) levels were detected by enzyme-linked immunosorbent assay(ELISA).Results The results showed that the secretion of IL-12 in LPS+morphine groups was significantly increased compared with blank control group and LPS control group(P0.01).But there was no difference between the LPS+morphine groups(P0.05).Conclusion Sustained morphine stimulation on the mouse bone marrow-derived dendritic cells can promote dendritic cells to mature.
