摘要 研究纤维素酶酶解-微波法提取黄芪多糖的新工艺.采用单因素分析与正交实验法,以黄芪多糖的提取率和质量分数为评价指标,考察酶料比、酶解时间、液固比、微波功率等工艺条件对多糖提取效果的影响.最佳工艺条件:酶料比57.6U/g,酶解时间60min,液固比10:1,微波功率480W,提取时间8min,重复提取1次,多糖提取率达到16.07%,多糖质量分数为88.40%,优于文献报道方法的提取效果.酶解-微波法具有耗时短、节约能源、提取率高、产物纯度较好等优点,是提取黄芪多糖的较理想方法. The microwave-assisted hydrolysis of cellulose was developed for the extraction of Astragalus Polysaccharides(APS). One-factor analysis and orthogonal experimental design were both applied to evaluate the extraction by the extraction rate and the purity of APS. The optimum enzymatic-microwave extraction condition was as follows: the liquid-to-solid ratio was 10 : 1; the enzyme ratio was 57.6 U/g; the cellulase reaction time was 60 min before 8 rain of microwave irradiation (480 W). The extraction was repeated once. In these conditions, the maximum extraction rate could reach 16.07% and the purity could be up to 88. 40%, which were much higher than those obtained from other extraction methods. The enzymatic-microwave assisted extraction is faster and more efficient with higher quality and therefore could be an ideal method for extracting the APS from Radix Astragali.
