免疫金银染色检测抗核抗体影响因素初探

STUDY OF FACTORS OF IN FLUENCE ON IGSS FOR ANA

对免疫金银染色检测抗核抗体时影响实验结果的一些因素进行了初步研究。认为控制好实验条件和银显影时间是关键。报道了不同显影温度、金标抗体工作浓度、银显影液中银盐含量、血清及金标抗体稀释液BSA含量、封闭液中BSA及羊血清含量等对银显影时间和染色结果的影响,及不同条件下银显影的最佳时间。

A methodological study of the influential factors in IGSS ( immunogold-silver staining ) for ANA ( antinuclear antibodies ) was performed, which showed that adequate length of developing time is a key to success.The factors that alter the developing time were found to include the amount of silver salt in developing fluid, the developing temperatnre the working concentration of GLA-Ig ( gold labeled anti-immunoglobulin ) ,the amount of BSA ( bovine serum albumin ) in the 2 fluids used for diluting sera and GIA-Ig, and the amounts of BSA and sheep serum in blocking fluids.The choices of time length for developing the end point under various methodological conditions were determined.
