摘要
目的研究盐酸头孢替安与血清白蛋白的结合作用,揭示盐酸头孢替安在体内的代谢过程。方法运用荧光光谱法测量盐酸头孢替安(cefotiam hydrochloride,CH)与牛血清白蛋白(bovine serum albumin,BSA)作用的荧光光谱,并运用Stern-Volmer方程及双对数方程对数据进行处理。结果盐酸头孢替安对BSA具有荧光猝灭作用,其作用机制为静态猝灭,根据Stern-Volmer方程及双对数方程求出了结合常数KA、结合位点数n及相关系数R。结论盐酸头孢替安与BSA形成了静态复合物,二者有弱结合位作用,这对于阐明盐酸头孢替安在体内的存储、转运和药理、毒理作用具有重要意义。
Objective To study the combination between cefotiam hydrochloride for injection(CH)and bovine serum albumin(BSA).Methods The interaction between CH and BSA was studied by fluorescence spectrum.The quenching data were analyzed in according to Stern-Volmer equation and double logarithmic equation.Results The fluorescence of BSA was quenched by CH,and the combination of them was a static quenching process.The binding constant and binding sites were obtained.Conclusion The combination between CH and BSA was weak.They had the weak force and formed a static compound.It was very important for its storage,transportation,pharmacological and toxicological effect in body by clarifying the interaction CH and BSA.
