摘要
目的:研究稀土离子Eu3+、Dy3+与槲皮素配合物的合成及抗肿瘤活性。方法:应用元素分析、摩尔电导、红外光谱、紫外光谱确定配合物的组成和结构,同时采用噻唑蓝(MTT)比色法测定配体及配合物对HepG2肝癌细胞株的抗肿瘤活性。结果:配合物的组成分别为EuC15H8O7C l10.6H2O和DyC15H8O7C l2.6H2O。槲皮素、铕-槲皮素和镝-槲皮素剂量依赖性地抑制HepG2细胞的增殖,且槲皮素稀土离子配合物对细胞增殖的抑制率显著高于槲皮素组。结论:配合物对HepG2肝癌细胞的抗肿瘤活性强于配体。
Objective:To investigate the synthetic method and antineoplasmic activity of the complex of rare earth element Eu(Ⅲ)、Dy(Ⅲ) with quercetin.Methods:On the basis of elemental analysis,molar conductance,IR spectra and UV spectra,the formula of the complex is given,moreover,MTT colorimetric assay was used to detect the anti-tumor activity of the ligand and the complex on human hepatocarcinoma cell line HepG2.Results: The formula of the complex is EuC15H8O7Cl10·6H2O and DyC15H8O7Cl2·6H2O.MTT assay showed that Quercetin,Eu-quercetin and Dy-quercetin dose dependently inhibited the proliferation of HepG2 cells,and the proliferation inhibition rates of cell treated with rare earth ion complexes of quercetin were significantly higher than those treated with quercetin.Conclusion:The anti-tumor activity of the complex is markedly stronger than that of the ligand.
