人参皂苷对人肺腺癌细胞转化生长因子β1表达的影响 被引量:2

摘要 目的探讨人参皂苷对人肺腺癌A549细胞及其对转化生长因子β1(TGF-β1)表达的影响。方法将人肺腺癌标准细胞株A549培养后完全随机分为人参皂苷组、对照组,采用四氮唑蓝法检测各组细胞增殖率;用酶联免疫吸附法测定各组细胞TGF—β1的表达水平,并分析TGF-β1的表达与A549细胞凋亡的相关性。结果当人参皂苷浓度为0.1、1、10、100μmoL/L时,A549细胞内TGF—β1表达水平逐渐降低,其吸光度(A)值分别为2.712±0.285、1.436±0.285、0.543±0.280、0.125±0.022,不同浓度的人参皂苷组间比较及其与对照组(A值为3.737±0.234)比较,差异均有统计学意义(均P〈0.05)。当人参皂苷浓度为1、10、100μmol/L时,A549细胞半数抑制浓度(IC50)分别为0.26±0.09、0.54±0.04和0.74±0.10,与阴性对照组(0.01±0.01)相比,差异均有统计学意义(均P〈0.05)。结论人参皂苷可以抑制肿瘤细胞TGF-β1的分泌,从而诱导肿瘤细胞凋亡。 Objective To investigate the role of ginsenoside Rg3 inducing apotosis in lung A549 cell line and the expression of transformin growth factor (TGF)-β1 levels. Methods Rg3-induced group and the control group were included in this study. 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) was used to determine the effect of Rg3 on apoptosis. The expression of TGF-β1 was analyzed by Enzyme-linked Immunosorbent Assay (ELISA). Results MTT assay showed that compared with the control group the apoptosis of Rg3-treated cells dramatically increased. Compared to the control group, the expression of TGF-β1 in the Rg3-treated decreased obviously. Conclusion Ginsenoside Rg3 has a high inhibition in the expression of TGF-β1 in A549 cell line and can induce the apotosis.
