鞘内注射Ro25-6981对大鼠慢性吗啡耐受的抑制作用研究

摘要 目的研究鞘内注射Ro25-6981对大鼠吗啡耐受的抑制作用及其可能机理。方法 SD大鼠40只,随机分成四组,每组10只。空白对照组(C1组):蛛网膜下腔注射0.9%生理盐水25μl;溶剂组(C2组):蛛网膜下腔注射5%二甲基亚矾(Dimethyl Sulfoxide,DMSO)溶剂25μl;鞘内注射组Ⅰ(R1组):蛛网膜下腔注射Ro25-6981注射液25μl(50μg);鞘内注射组Ⅱ(R2组):蛛网膜下腔注射Ro25-6981注射液25μl(100μg)。四组大鼠完成鞘内注射后,参照文献[3]方法分别建立慢性吗啡耐受模型。注射后第1天、第3天、第5天和第7天采用热水甩尾法(hot-water tail-flick)测定甩尾潜伏期(Tail Flick Latency,TFL)的变化来评价吗啡的镇痛效果,同时采用斜板实验方法进行运动功能检测。结果同C1组比较:R1组第3天和第5天MPE%差异有统计学意义(p<0.05),R2组各时间点MPE%的差异有统计学差异(p<0.05),C1和C2组差异无统计学意义。与第1天比较:R1组第5天和第7天MPE%逐渐降低(p<0.05),第3天变化不明显;R2组第7天MPE%明显降低(p<0.05),第3天和第5天变化不明显(p>0.05)。不同时间点各组大鼠斜板角度的变化差异无统计学意义(p>0.05)。结论 Ro25-6981鞘内注射能够在一定程度上抑制大鼠慢性吗啡耐受的形成且不影响运动功能,表明NR2B在吗啡耐受形成中起了关键的作用。
