摘要
目的评价ATP的感性钾通道(KATP)开放剂吡那地尔对大鼠慢性压力超负荷性左心室肥厚/心力衰竭模型动物心功能的影响。方法将成年Wistar大鼠60只随机分为3组:手术药物组(A组)、手术对照组(B组)和假手术对照组(C组),A组大鼠从术后4周开始应用吡那地尔,比较术后第12周和16周B型钠尿肽(BNP)及血流动力学监测指标的变化。结果腹主动脉缩窄术后12周,B组左心室收缩末压(LVSP)显著升高(P<0.05),A组LVSP值明显低于B组(P<0.01),术后16周时低于C组(P<0.05)。A、B组+dP/dt明显低于C组(P<0.01),术后16周A组+dP/dt略有回升。A、B组-dP/dt值在术后均明显低于C组(P<0.01),A组略高于B组(P>0.05)。B组BNP在腹主动脉缩窄术后均明显高于C组(P<0.01),A组BNP水平在术后12周明显高于C组(P<0.05),第16周与C组相当(P>0.05),均低于同期的B组(P<0.05或<0.01)。结论吡那地尔可显著改善慢性压力超负荷性心力衰竭模型大鼠的心功能。
Objective To investigate the effects of pinacidil on the cardiac function in different time points in rat models with chronic heart failure. Methods Sixty Wistar rats were randomly divided into three groups :group A (aortic banding group plus pinacidil), group B (aortic handing group) and group C (shamoperation group). Pinacidil was used at 4 weeks after the operation, and BNP concentration and hymodanamic parameters were detected for the three groups at 12 week and 16 week after the operation. Results The LVSP in group B was increased at 12 weeks after operation ( P 〈0. 01 ) ,and LVSP in group A was significantly lower than that of group B( P 〈0.01) ,which was lower than that in group C in 16 weeks ( P 〈0.05). The + dp/dt in group A and group B was significantly lower than that of group C after the operation ( PP 〈 0.01 ) ,and the + dp/ dt in group A was increased,as compared with that before 16 weeks. The + dp/dt in group A and group B was obviously lower than that in group C after operation ( P 〈 0.01 ), and dp/dt in group A was significantly higher than that of group B ( P 〉 0.05 ). The BNP in group B was significantly higher than that in group C after operation ( P 〈0.01) ,and BNP in group A was obviously higher than that in group C at 12 weeks after operation ( P 〈 0.01 ) ,which was similar to that in group C at 16 weeks ( P 〉0.05) ,however,which was significantly lower than that in group B( 12 week: P 〈 0.01;16 week: P 〈 0.01 ). Conclusion The pinacidil can significantly improve the cardiac function of rat models of heart failure with chronic pressure overload.
