摘要
研究了饥饿24h和48h对多刺裸腹溞Moina macrocopa糖和蛋白质代谢中间代谢产物以及主要代谢酶的影响。结果显示,饥饿能够显著影响多刺裸腹溞机体内葡萄糖、丙酮酸、总可溶性蛋白和总氨基酸含量;同时对琥珀酸脱氢酶(SDH)、丙酮酸激酶(PK)、谷丙转氨酶(GPT)和谷草转氨酶(GOT)的活力也有不同程度的影响。饥饿24h和48h,葡萄糖含量分别降低了32.17%和37.55%;丙酮酸含量分别降低了46.04%和38.53%,与对照组相比,发生了显著性变化(P<0.05)。饥饿24h可溶性蛋白含量降低了32.14%(P<0.05);饥饿24h和48h总氨基酸含量呈极显著下降趋势(P<0.01),分别下降了61.47%和65.38%。实验证明,饥饿胁迫可诱导SDH活力增加,48h时活力极显著升高(P<0.01);而PK活力变化不显著(P>0.05);GPT和GOT两种转氨酶活性在饥饿胁迫下呈上升趋势,但是与对照组相比无显著性差异(P>0.05)。
The effect of short-term starvation(24 h and 48 h) on carbohydrate and protein metabolism of Moina macrocopa was investigated.This experiment was conditioned at(25±1)℃ in water temperature,pH at 7.2 and dissolved oxygen at 6.0 mg/L.Several key enzyme activities of carbohydrate and protein metabolism were studied including pyruvate kinase(PK),succinate dehydrogenase(SDH),glutamic oxaloacetic transaminase(GOT),glutamic pyruvic transaminase(GPT) and glucose,pyruvate.Total soluble protein content and total amino acids content were also quantified.The results showed that glucose and pyruvate content decreased rapidly as the starvation extended.Glucose content decreased significantly by 32.17% and 37.55% compared with control after 24 h and 48 h of starvation.After 24 h of starvation,pyruvate content also decreased significantly,and it almost fell to 50% of control.Compared with control,the soluble protein level was significantly decreased(P0.05) by 32.14% under starvation for 24 h.The total amino acids content was remarkably reduced(P0.01) by 61.47% and 65.38% respectively after starvation of 24 h and 48 h.In response to the starvation,the activities of SDH increased by more than 50% at 48 h of the starvation compared with control.However,no significant changes of PK,GOT and GPT activities were observed.
