大鼠肝癌演进中凝血因子Ⅴ的可变剪接

摘要 为得到未知差异表达基因EST-5全长及探讨其在SD大鼠肝癌演进中的作用,采用cDNA末端快速扩增技术(rapid amplification of cDNA ends,RACE),扩增5'端和3'端序列,PCR产物TA克隆后测序,BLAST检索分析所得序列。结果显示,该序列与挪威大鼠coagulation factor Ⅴ(Rattus norvegicus coagulation factorⅤ,FⅤ)序列(Gen-Bank登录号:NM_001047878)相似性高达98%,且EST-5以插入的形式存在于该序列中,推测大鼠肝癌演进过程中,凝血因子Ⅴ可能存在另一种可变剪接的形式。 To get the full-length sequence of the differentially expressed gene EST-5 and explore its role in primary rat hepatic cancer,rapid amplification of cDNA ends(RACE) technology was used to amplify 5' and 3' end sequences,then cloned the gene for sequencing by TA cloning technique.The sequence was analyzed by BLAST.The results showed that its sequence had a high similarity(up to 98%) with NM-001047878 Norway rat coagulation factor Ⅴ.Due to the insertion of EST-5 into the sequence,coagulation factor Ⅴ may be suggested to have another alternative splicing form in primary rat hepatic cancer.
