摘要
目的本研究旨在用TCA-丙酮沉淀法对聚合草叶蛋白进行提取,为聚合草叶蛋白的大规模制备奠定基础。方法测定聚合草干叶、鲜叶中的蛋白含量,提取的蛋白经真空冷冻干燥后称重,确定所提蛋白的纯度并计算蛋白提取率。结果以鲜叶为原料时的提取率为51.76%,所得蛋白的纯度为54.2%;以干叶为原料时,蛋白提取率为46.45%,蛋白纯度为48.3%。结论利用TCA-丙酮沉淀法从聚合草鲜叶中提取粗蛋白优于干叶。
Objective To study the extraction of leaf protein of Symphytum Pezegrinum L.with TCA-acetone precipitation so as to lay the foundation for the large-scale preparation of Symphytum Pezegrinum L.Methods Detecting the protein contents in dry leaf and fresh leaf of Symphytum Pezegrinum L.,weighting the extracted protein after vacuum freezing-drying and determining the purity of the extracted protein and calculating protein extraction rate.Results The extraction rate of fresh leaf was 51.76% and the protein purity 54.2%.The extraction rate of dry leaf was 46.45% and the purity 48.3%.Conclusion The extraction of crude protein from fresh leaf of Symphytum Pezegrinum L.with TCA-acetone precipitation is superior to that from dry leaf.
