摘要 用改良的CTAB法提取15份果桑无性系的基因组DNA,通过RAPD分子标记技术,构建不同果桑无性系的指纹图谱。结果表明:每条引物扩增出8~15条清晰地条带,其中多态性条带数为4~15条,多态性比例为50.0%~100.0%。20条随机引物共得到232条清晰的条带,多态性条带为178条,平均多态性比例为76.7%;利用随机引物OPA-09和OPD-05的扩增产物电泳图为基础构建的指纹图谱可以鉴定出15个果桑无性系。 Modified CTAB method was clones. The RAPD fingerprints of 15 adopted to extract total DNA of 15 fruit mulberry (Morus spp. ) fruit mulberry clones were constructed. The results showed that 8-15 bands were clearly amplified from each primers, of which, 4-15 bands were of potymorphism and the percentages of primers with polymorphism were from 50.0% to 100.0%. Totally 232 bands were produced using 20 random primers, in which 178 bands showed polymorphism. Fifteen fruit mulberry clones can be identified by using the fingerprints established on the electrophoresis map of products amplified by primers of OPA-09 and OPD-05.
