邻苯二甲酸二(2-乙基己基)酯宫内暴露所致子代雄性大鼠睾丸氧化应激及维生素C的干预作用 被引量:5

Effect of Exposure to Di-2-ethylhexyl Phthalate during Pregnancy on Testis Lipid Peroxidation of Offspring Rats and Protective Effect of Vitamin C

摘要 目的探讨邻苯二甲酸二(2-乙基己基)酯(di-2-ethylhexyl phthalate,DEHP)宫内暴露对子代雄性大鼠睾丸脂质过氧化水平的影响及维生素C(vitamin C,Vit C)的抗氧化应激作用。方法将20只健康成年清洁级Wistar妊娠大鼠按体重随机分为6组,分别为对照(玉米油)组和低(10 mg/kg)、中(100 mg/kg)、高(500 mg/kg)剂量DEHP染毒组以及中(100mg/kg)、高(500 mg/kg)剂量DEHP+Vit C染毒组,中、高剂量DEHP染毒组每组孕鼠4只,其余各组每组孕鼠3只。自妊娠第0~19天,灌胃染毒DEHP,染毒容量为10 ml/kg,每天1次;采取自由饮水方式染毒Vit C(125 g/L),每天约40 ml。子代雄性大鼠出生后第6周,测定睾丸组织中脂质过氧化产物丙二醛(MDA)、还原型谷胱甘肽(GSH)含量和过氧化氢酶(CAT)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)的活力。结果与对照组相比,高剂量DEHP染毒组仔鼠睾丸组织中MDA含量明显升高,高剂量DEHP染毒组和高剂量DEHP+Vit C染毒组GSH-Px活力以及高剂量DEHP染毒组和中、高剂量DEHP+Vit C染毒组SOD活力明显下降,差异均有统计学意义(P<0.05)。各染毒组仔鼠睾丸组织中GSH含量和CAT活力与对照组比较,差异均无统计学意义。与中剂量DEHP单独染毒组相比,中剂量DEHP+Vit C染毒组仔鼠睾丸组织中GSH含量和SOD、CAT活力均升高,但仅SOD活力明显升高,差异具有统计学意义(P<0.05)。与高剂量DEHP单独染毒组相比,高剂量DEHP+Vit C染毒组仔鼠睾丸组织中MDA含量降低,GSH含量、GSH-Px活力及CAT活力升高,但差异均无统计学意义。结论 DEHP宫内暴露可导致子代雄性大鼠睾丸组织的脂质过氧化水平增加,Vit C对于DEHP所致生殖系统的氧化损伤具有一定的保护作用。 Objective To study the effect of exposure to di-2-ethylhexyl phthalate (DEHP) during pregnancy on lipid peroxidation in the testis of offspring rats and the protective effect of Vit C. Methods Twenty pregnant Wistar rats were divided into 6 groups, control group (corn oil), 10 mg/kg DEHP group, 100 mg/kg DEHP grrup, 500 mg/kg DEHP, 100 mg/kg DEHP+125 g/L Vit C and 500 mg/kg DEHP+ 125 g/L Vit C. The treatment was conducted from GD 0 to 19, for DEHP through gavage, once a day, and for Vit C through drinking water. At the 6th week of life,the testis of all offspring rats were removed immediately, and the organ indices were measured, the level of MDA was detected by TBA method, the levels of the GSH and GSH-Px were analyzed by modified DTNB method, the activity of the SOD was measured by xanthine oxidase method, the activity of the CAT was observed by spectrophotometry. Results Compared with the control,the level of MDA in 500 mg/kg DEHP group increased significantly (P〈0.05). The levels of GSH-Px significantly decreased in 500 mg/kg DEHP and in 500 mg/kg DEHP+Vit C group, Vit C presented an obvious protective effect. The activity of the SOD decreased significantly in 100 mg/kg DEHP,500 mg/kg DEHP,and in 100 mg/kg DEHP+Vit C group, Vit C also presented an obvious protective effect. Conclusion Exposure to DEHP during pregnancy may cause the increase of lipid peroxidation level in the testis of offspring in rats. Vitamin C has a protective effect on DEHP induced oxidative damage up to a point.
