摘要
目的:研究表皮生长因子(EGF)对体外培养的大鼠牙囊细胞增殖活性的影响。方法:原代培养Wistar大鼠牙囊细胞,选择生长良好的第3代细胞,分别加入不同浓度的EGF(0.5、1、5、10、50 ng/mL)与牙囊细胞共孵育5 d,MTT法对比观察不同浓度的EGF对牙囊细胞增殖活性的影响。数据采用SPSS 13.0软件包进行方差分析。结果:在EGF浓度为5~10 ng/mL时,牙囊细胞的增殖活性显著增高(P<0.05),10 ng/mL时达到最高(P<0.01)。结论:EGF可作用于牙囊细胞,合适浓度的EGF能显著增加牙囊细胞的增殖活性。
Objective:To study the effects of epidermal growth factor(EGF) on the proliferation of rat dental follicle cells.Method:Dental follicle was harvested from 4~6 days postnatal Wistar rat and cultured in α-MEM with 15 % FBS.Cell origin was identified by the expression of Vimentin and Cytokeratin.The cells of the third passage were used.Controls were then cultured in α-MEM containing 0.5 % serum,experimental cultures were grown in this plus different concentrations(0.5,1,5,10,50 ng/mL) of epidermal growth factor in medium for 5 days.MTT assays were used to evaluate the proliferation of the control and experimental groups.The results were analyzed by SPSS 13.0 software package.Result:The typical characterization of cultured cells was plemorphism and there were two major cell types.The results of immunocytochemistry indicated that dental follicle cells expressed Vimentin positively while the expression of Cytokeratin was negative,which indicated that the cells were originated from mesenchyme.Epidermal growth factor at the concentration of 5~10 ng/mL significantly stimulated the proliferation of the dental follicle cells(P 0.05),and the prime effective concentration of EGF on dental follicle cells was 10 ng/mL.Conclusion:Proper concentrations of EGF may promote dental follicle cell proliferation.
