核仁素在骨肉瘤组织中的表达及其意义

Study on the expression and significance of nucleolin in osteosarcoma

摘要 目的探讨骨肉瘤中核仁素蛋白的表达情况,明确其表达与骨肉瘤临床病理遗传的关系。方法骨肉瘤组织35例,骨软骨瘤组织15例,应用免疫组化技术,检测石蜡包埋组织中核仁素蛋白的表达,并分析核仁素蛋白的表达与骨肉瘤临床病理遗传的关系。结果(1)核仁素蛋白在骨肉瘤组织中的阴性、弱、中、强表达依次为8.6%、20%、34.3%、37.1%,与骨软骨瘤组织(分别为40%,46.7%,13.3%,0)相比,其表达水平显著上调(P〈0.01)。(2)核仁素蛋白在普通型骨肉瘤中阴性、弱、中、强表达依次为6.9%、13.8%、34.5%、44.8%,与骨旁骨肉瘤(分别为16.7%,50%,33.3%,0)相比,其表达水平上调(P〈0.05)。(3)核仁素蛋白在成骨型骨肉瘤中阴性、弱、中、强表达依次为5.9%、5.9%、29.4%、58.8%,成软骨型骨肉瘤中分别为0%、33.3%、33.3%、33.3%,成纤维型骨肉瘤中分别为16.7%、16.7%、50%、16.7%,不同亚型普通骨肉瘤组织中,核仁素蛋白的表达差异无统计学意义(P〉0.05)。结论核仁素蛋白的表达与骨组织病变的性质有关,其表达水平与骨肉瘤的病理遗传有关。 Objective To investigate the expression of nucleolin in osteosarcoma and its clinical significance. Methods The expression of nucleolin was evaluated by immunohistochemistry in paraffin-em- bedded tissues from 35 cases of osteosarcoma and 15 cases of osteoehondroma. Results Among the osteo- sarcoma cases, 8.6% were negative for nucleolin, 20% weakly positive, 34. 3% moderate positive, 37.1% strong positive. The positive rate of nucleolin in osteochondroma was 40% , 46.7% , 13.3% , 0%0 , respectively. The nucleolin expression showed significant difference in the osteosarcoma and osteochondroma ( P 〈 0. 01 ). Among the conventional osteosarcoma cases, 6. 9% were negative for nucleolin, 13.8% weakly positive, 34. 5 % moderate positive, 44. 8 % strong positive. The positive rate of nueleolin in paros- teal osteochondroma was 16. 7% , 50% , 33.3% , 0% , respectively. The nucleolin expression showed re- markable correlation with pathological and genetical classification ( P 〈 0. 05 ). The positive rate of nucleo- lin in osteoblastic osteochondroma was 5.9%, 5.9%, 29. 4% , 58. 8% , while in chondroblastic osteoehon- droma 0% , 33.3% , 33.3%, 33.3% and 16. 7% , 16. 7% , 50%, 16.7% in fibroblastic osteochondroma, respectively. The positive expressions of nucleolin were almost the same in each subtype of osteosarcoma ( P 〉 0.05 ). Conclusions The nucleolin expression had correlation with property of tumors in bone, and the positive expressions of nueleolin had correlation with pathological and genetic classification of osteosarcoma.
