摘要
通过单因素实验、正交实验研究了不同碳源、氮源、碳氮比例、缓冲盐浓度、微量元素及其他生长因子对瑞士乳杆菌(Lactobacillushelveticus H9)增殖培养的影响。并采用响应面法对优选的碳源、氮源和缓冲盐类的组成质量浓度进行优化,得到的增殖培养基:葡萄糖20 g/L,大豆蛋白胨10 g/L,酵母粉5 g/L,NaAC为7 g/L,K2HPO4为3 g/L,柠檬酸钠1 g/L,MgSO4.7H2O为200 mg/L、MnSO4.5H2O为81 mg/L、L-半胱氨酸0.25 g/L、吐温80为1 g/L。Lactobacillus helveticus H9在此增殖培养基中经37℃,10 h培养活菌数可达到1.34×1010mL-1,比在普通MRS中提高近35倍。
Lactobacillus helveticus H9 was isolated from Kurut in Luoma of Tibet.The effects of different carbon source, nitrogen source, proportion of carbon and nitrogen source, buffer salts and microelement on the growth of Lactobacillus helveticus H9 were studied through single factor test and orthogonal experimental design. The enrichment medium for Lactobacillus helveticus H9 was optimized by response surface methodology and the composition was consist of ghicose 21.0 g/L, soy peptone 10.0 g/L, yeast extract 5.0 g/L, K2HPO4 2.929 g/L, sodium acetate 6.835 g/L, sodium citrate 0.976 g/L, MgSO4· 7H2O 200 rag/L, MnSO4·5H2O 81 mg/L, L-cysteine0.25 g/L, tween-80 1.0 g/L. After cultivated in enrichment medium for 10 h at 37 ℃, the living cells of Lactobacillus helveticus H9 were 1.34×10^10 mL^-1, which was about 35 times higher than in MRS.
