积雪草苷对脊髓损伤大鼠的保护作用及机制研究 被引量:5

摘要 目的探讨积雪草苷(AS)对大鼠脊髓损伤后星形胶质细胞保护作用及D-丝氨酸表达的影响。方法成年大鼠于脊髓损伤术前30 min给药,损伤后不同时间点(1,3,7,14,28 d)处死。用免疫组化染色观察损伤脊髓组织星形胶质细胞胶质纤维酸性蛋白(GFAP)、胶质源性神经营养因子(GDNF)及神经元特异性烯醇化酶(NSE)的表达变化,化学发光法测定损伤脊髓段D-丝氨酸含量的变化。结果与阴性对照组相比,积雪草苷组可明显提高GFAP、GDNF及NSE的表达(P<0.05),积雪草苷组D-氨基酸表达明显减少(P<0.05)。结论积雪草苷能使星形胶质细胞损伤减小,增强GFAP、GDNF及NSE的表达,抑制大鼠脊髓损伤D-丝氨酸表达,从而保护神经元细胞,使NSE表达增加。 Objective To investigate the effects of asiaticoside on astrocytes and on D-serine expression in rats after spinal cord injury(SCI). Methods The rat model of impact injury of spinal cord(T8-T10) was made by Allen's method. Adult SParuge-Dawley(SD) rats were administered the corresponding medicine 30 min before the injury, and then were executed at day 1, 3, 7, 14 and 28. We measured the expression levels of glial fibrillary acidic protein (GFAP) , glial cell line-derived neurotrophic factor(GDNF), and neuronspecific enolase (NSE) in the astrocytes by immunohistochemical assay, and observed the change of D-serine content in the injured spinal cord by using the method of chemiluminescence. Results Compared with the negative control group, the expression levels of GFAP, GDNF and NSE were increased, and D-serine expression was decreased in asiaticoside groups(P 〈 0.05). Conclusion The protective effect of asiaticoside against astrocytes is probably related to the increase of GFAP, GDNF and NSE expression and to the decrease of D-serine expression in rats with spinal cord injury.
